Hello, Medusa Dracul! (Great name!)
Well, once I have at least a vague idea of the plot, I usually decide on general traits such as dependable, strong willed, artistic, spunky, etc... Then I think about what's important to the character to drive their motivations and decisions. For example, Riley's family was very important to her in In the Band, which lead to several plot turns. Lastly, I try to give each character some individuality with 'props.' These could be words they use a lot, interests they have, certain clothes they wear, etc...
However, I will admit that I never totally feel I know a character until I reach about fifty pages, and sometimes I do back track when they become fully rounded to me.

Hi Mary!
She does show up in all the sequels. More so in Ink My Heart than With the Band. The last one, which I'm working on, has a lot of Riley in it. She not only mentors Gabe, the new drummer, and is friends with his love interest, but her new band is somewhat part of the plot--not a spoiler since they play in chapter one. Unless things change...
